Abstract:

To determine the role of endothelin-1 (ET-1) and its receptors in the regulation of calcitonin gene-related peptide (CGRP) release, male Wistar rats were divided into six groups and subjected to the following treatments for 1 wk with or without ABT-627 (an ETA receptor antagonist, 5 mg·kg−1·day−1 in drinking water) or A-192621 (an ETB-receptor antagonist, 30 mg·kg−1·day−1 by oral gavage): control (Con), ET-1 (5 ng·kg−1·min−1 iv), Con + ABT-627, Con + A-192621, ET-1 + ABT-627, and ET-1 + A-192621. Baseline mean arterial pressure (MAP, mmHg) was higher ( P < 0.05) in Con + A-192621 (122 ± 4) and ET-1 + A-192621 (119 ± 4) groups compared with Con (104 ± 6), ET1 (106 ± 3), Con + ABT-627 (104 ± 3), and ET1 + ABT-627 (100 ± 3) groups. Intravenous administration of CGRP8–37 (a CGRP receptor antagonist, 1 mg/kg) increased MAP ( P < 0.05) in ET-1 (13 ± 1), Con + A-192621 (12 ± 1), and ET-1 + A-192621 (15 ± 3) groups compared with Con (4 ± 1), Con-ABT-627 (4 ± 1), and ET-1 + ABT-627 (5 ± 1) groups. Plasma CGRP levels (in pg/ml) were increased ( P < 0.05) in ET-1 (57.5 ± 6.1), Con + A-192621 (53.9 ± 3.4), and ET-1 + A-192621 (60.4 ± 3.0) groups compared with Con (40.4 ± 1.6), Con + ABT-627 (40.0 ± 2.9), and ET-1 + ABT-627 (42.6 ± 1.9) groups. Plasma ET-1 levels (in pg/ml) were higher ( P < 0.05) in ET-1 (2.8 ± 0.2), ET-1 + ABT-627 (3.2 ± 0.4), Con + A-192621 (3.3 ± 0.4), and ET-1 + A-192621 (4.6 ± 0.3) groups compared with Con (1.1 ± 0.2) and Con-ABT-627 (1.3 ± 0.2) groups. Therefore, our data show that ET-1 infusion leads to increased CGRP release via activation of the ETA receptor, which plays a compensatory role in preventing ET-1-induced elevation in blood pressure.

The structures of Ni(C≡CR)(PPh3)(η-C5H5) (R = Ph (1), C6H4-4-NO2 (2), 4-C6H4C6H4-4′-NO2 (3), (E)-4-C6H4CH=CHC6H4-4′-NO2 (4), 4-C6H4C≡CC6H4-4′-NO2 (5), 4-C6H4N=CHC6H4-4′-NO2 (6)) have been determined by single-crystal X-ray diffraction studies, refining by full-matrix least-squares analysis. Abstract:

Background Death associated with coronary heart disease (CHD) depends in part on the time since the myocardial infarction (MI) and modification of risk factors. Methods This observational, retrospective 4-year follow-up study consisted of 804 patients (628 men). The participants completed a questionnaire reporting diet, demographic factors, personal behavior (smoking, physical activity), anthropometry, prior medical conditions (hypertension, diabetes mellitus), and recent medication. Results During 48 months of follow-up, 12% of men and 15% of women died. Older age, longer duration of smoking, and frequency of exercise were significantly different between survivors and the deceased ( P = .014, P = .014, P = .001, respectively). Multivariate analysis revealed associations with years of smoking (odds ratio, OR: 1.10, P = .025), treatment with nitrates (OR: 4.81, P = .024), and increased frequency of exercise (OR: 0.42, P = .013), adjusting for age and gender. Conclusions We should emphasize cessation of smoking and increased physical activity in MI survivors. Antismoking programs should start at an early age.

Abstract:

High intensity interval training is frequently implemented using the 4 × 4 protocol where four 4-min bouts are performed at heart rate (HR) between 85 and 95% HR max. This study identified the HR and power output response to the 4 × 4 protocol in 39 active men and women (age and VO2 max = 26.0 ± 6.1 years and 37.0 ± 5.4 mL/kg/min). Initially, participants completed incremental cycling to assess VO2 max, HR max, and peak power output (PPO). They subsequently completed the 4 × 4 protocol, during which HR and power output were monitored. Data showed that 12.9 ± 0.4 min of 16 min were spent between 85 and 95% HR max, with time spent significantly lower in interval 1 (2.7 ± 0.6 min) versus intervals 2–4 (3.4 ± 0.4 min, 3.4 ± 0.3 min, and 3.5 ± 0.3 min, d = 2.4–2.7). Power output was highest in interval 1 (75% PPO) and significantly declined in intervals 2–4 (63 to 54% PPO, d = 0.7–1.0). To enhance time spent between 85 and 95% HR max for persons with higher fitness, we recommend immediate allocation of supramaximal intensities in interval one.

Abstract:

Four copolymers (P(tCz (tris(4-carbazoyl-9-ylphenyl)amine)-co-bTP (2,2'-bithiophene)), P(tCz-co-CPDT (4H-cyclopenta[2,1-b:3,4-b’]dithiophene)), P(tCz-co-DTC (3,6-di(2-thienyl)carbazole)), and P(tCz-co-CPDTK (cyclopentadithiophene ketone))) are deposited on indium tin oxide (ITO) surfaces using electrochemical polymerization. Spectroelectrochemical properties of copolymer electrodes reveal that the colors of P(tCz-co-bTP) film are pinkish-orange, light olive green, light grayish blue, and dark blue at 0.0, 0.8, 1.2, and 1.6 V, respectively, whereas the color variations of P(tCz-co-CPDTK) film are light yellow, yellow, and blue at 0.0 V, 0.8 V, and 1.5 V, respectively. The ΔT of P(tCz-co-bTP), P(tCz-co-CPDT), P(tCz-co-DTC), and P(tCz-co-CPDTK) films are estimated to be 43.0% at 967 nm, 28.7% at 864 nm, 43.6% at 870 nm, and 24.5% at 984 nm, respectively. Five electrochromic devices (ECDs) are assembled using the tCz-based hom*opolymer and copolymers as coloring electrodes, and poly(2,2-dimethyl-3,4-propylenedioxythiophene) (PProDOT-Me2) as the complementary electrode. P(tCz-co-DTC)/PProDOT-Me2 ECD reveals high transmittance change (45.9% at 624 nm), P(tCz-co-CPDTK)/PProDOT-Me2 ECD shows high η (513.0 cm2 C−1 at 582 nm), and P(tCz-co-bTP)/PProDOT-Me2 ECD presents short switching time (less than 0.4 s) at 628 nm. Moreover, these ECDs show satisfactory redox stability and open circuit stability.

Abstract:

This study was designed to quantify the long-term contribution of endogenous endothelin-1 (ET-1) and ETA receptors to the regulation of arterial pressure under normal conditions in nonhuman primates. Therefore, mean arterial pressure (MAP) and heart rate were measured 24 h/day with the use of telemetry techniques in conscious cynomolgus monkeys under control conditions, during administration of an ETA selective receptor antagonist (ABT-627; 5 mg/kg, 2 times a day by mouth, 4 days), and a 6-day posttreatment period. Systemic ETA blockade reduced MAP (24 h) from 89 ± 3 to 82 ± 2 and 79 ± 2 mmHg on days 1 and 4, respectively. Subsequently, MAP remained suppressed for 3 days posttreatment. Heart rate increased from 111 ± 5 to 122 ± 4 and 128 ± 6 beats/min on days 1and 4 of ABT-627, respectively, and remained above control for 3 days posttreatment. Plasma ET-1 concentration increased from 1.0 ± 0.3 to 1.9 ± 0.4 pg/ml in response to ABT-627 ( day 4) but decreased to control values 4 days posttreatment. These data demonstrate a physiologically important role for endogenous ET-1 and ETA receptors in the long-term regulation of arterial pressure and plasma ET-1 levels in the conscious nonhuman primate.

Abstract:

The synthesis of 17α-acetoxy-3β-[(β-D-glucopyranosyl)oxy]-6α-methylpregn-4-en-20-one, the glucoside of medroxyprogesterone acetate (MPA-glu), is described. MPA-glu and 14-amino-20β-hydroxy-3β-[(α-L-rhamnopyranosyl)oxy]-5β,14β-pregnane (LND 623), pregnane glycosides that bind to the digitalis receptor, and digoxin, a cardiac glycoside, were infused intravenously into the anesthetized guinea pig. Each of the three steroids significantly enhanced urinary volume and sodium excretion without affecting blood pressure and creatinine clearance. Potassium excretion was markedly enhanced by digoxin but unaffected by MPA-glu or LND 623. These observations conform to previous work that demonstrated, in the rat, potassium-sparing diuresis by the glucoside of 14β-hydroxyprogesterone, a cardiotonic pregnane. There is a dissociation between potency to inhibit [3H]ouabain binding and the extra ATPase actions of the digitaloid pregnanes.Key words: pregnanes, cardiac glycosides, diuretic, LND 623, medroxyprogesterone acetate.

Abstract:

Abstract Objectives Determine the effect of three 100% black bean pastas made with standard (Knife Mill) and novel flour milling techniques (sonic wave) on postprandial glycemic response in healthy normoglycemic adults in comparison to whole black beans and a white bread control. Methods Heat treated black beans (Zenith) were milled into flour using a commercial knife mill or a sonic wave mill. The latter generated bean flours with low (cyclone), and medium (combined) protein concentrations from which pastas of 100% black beans were made commercially. Fifteen adults (6 men, 9 women, mean age 23.4 ±2.7 y; nonsmokers; BMI &lt;30) were recruited for a randomized 5 × 5 cross-over trial. The test meals were white bread control (no sauce), whole black beans and 3 black bean pastas served with a standard amount of spaghetti sauce to net 50 grams of available carbohydrate (CHO). The test meals were similar in macronutrient content except for protein and fiber (knife mill protein 19.5, fat 6.3, fiber 22; combined 20.5, 7.5, 22.4; cyclone 13.7, 6.1, 19.5; whole beans 20.9; 5.4, 28.2). Fasted participants consumed a test meal within 7–10 minutes. Venous blood draws were taken at 30, 60, 90, 150, and 180 minutes post meal start. Glucose was analyzed by a professional laboratory (Quest Diagnostics, Chicago, IL). Results Net glucose response based on the incremental area under the curve (iAUC) for 0–180 minutes was significantly different by treatment for all four test meals in comparison to the white bread control (P = 0.002). For the 0–60 iAUC increment, participants showed a lower response to whole black beans than white bread (P = .000), knife mill (P = .024), combination (P = .024), or cyclone (P = .014) pastas. The 0–120 iAUC period showed significantly higher net glucose for the white bread than the 4 bean treatments. Black beans were significantly different from the knife mill (P = .033) and cyclone pasta formulations (P = .049). Conclusions Black bean pastas improve glycemic response regardless of milling type in relation to a control food providing evidence that has promising implications for dietary guidance to reduce postprandial glucose and related health risks through processed food consumption. Abstract:

ABSTRACT Aim To evaluate pH and antibacterial activity of pastes with calcium hydroxide [Ca(OH)2] and zinc oxide (ZnO) microparticles (micro) or nanoparticles (nano) and association with 0.4% chlorhexidine against Enterococcus faecalis. Materials and methods The following pastes were analyzed: Ca(OH)2/ZnO micro, (2) Ca(OH)2/ZnO nano, (3) Ca(OH)2/ ZnO micro + 0.4% chlorhexidine, (4) Ca(OH)2/ZnO nano + 0.4% chlorhexidine. Antibacterial activity against E. faecalis was evaluated by agar diffusion test. The direct contact test on planktonic cells of E. faecalis was performed for 30 and 60 seconds. Root canals from bovine teeth were filled with the pastes and pH was evaluated after 1, 7, 14, 21, 30 and 60 days. The data obtained were submitted to the statistical tests analysis of variance (ANOVA) and Tukey or Kruskal-Wallis and Dunn test, with a 5% significance level. Results Calcium hydroxide and zinc oxide nano, and the pastes with 0.4% chlorhexidine were more effective in agar diffusion test. In the direct contact test, the pastes with chlorhexidine showed the highest effect after 30 seconds. All pastes eliminated E. faecalis after 60 seconds. All pastes promoted an increase in pH. The highest increase in pH was observed with nanoparticle medications after 1 and 7 days (p < 0.05). After this period, the pastes presented similar pH increase. Conclusion It was concluded that calcium hydroxide and zinc oxide nanoparticles promoted greater initial alkalinization. The antimicrobial activity of the pastes against E. faecalis is favored by the association with chlorhexidine. Clinical significance Although nanoparticles of calcium hydroxide and zinc oxide promoted antibacterial effect, the activity against E. faecalis is favored by association with chlorhexidine. Abstract:

AbstractKitagohaite, ideally Pt7Cu, is a new mineral from the Lubero region of North Kivu, Democratic Republic of the Congo. The mineral occurs as alluvial grains that were recovered together with other Pt-rich intermetallic compounds and Au. Kitagohaite is opaque, greyish white and malleable and has a metallic lustre and a grey streak. In reflected light, kitagohaite is white and isotropic. The crystal structure of kitagohaite is cubic, space group Fmm, of the Ca7Ge type, with a = 7.7891(3) Å, V = 472.57(5) Å3 and Z = 4. The strongest diffraction lines [d in Å(I)(hkl)] are: 2.246 (100)(222), 1.948(8)(004), 1.377 (77)(044), 1.174(27)(622), 1.123 (31)(444) and 0.893 (13)(662). The Vickers hardness is 217 kg mm−2 (VHN100), which is equivalent to a Mohs hardness of 3½ and the calculated density is 19.958(2) g cm−3. Electron-microprobe analyses gave a mean value (n = 13) of 95.49 wt.% Pt and 4.78 wt.%Cu, which corresponds to Pt6.93Cu1.07 on the basis of eight atoms. The new mineral is named for the Kitagoha river, in the Lubero region.

The syntheses and room-temperature single-crystal X-ray structural characterization of binuclear 1 : 2 adducts formed between silver(I) (pseudo-)halides, AgX, and triphenylarsine, AsPh3, for X = Cl, Br, I, SCN (1)–(4), are described. The chloride (1), obtained from 2-methylpyridine, is triclinic, P-1, a 10·410(2), b 12·716(2), c 14·196(6) Å, α 113·38(2), β 109·41(2), γ 75·08(1)°, Z = 1 (dimer); conventional R on F was 0·037 for No 3979 independent ‘observed’ (I > 3σ(I)) reflections. The bromide (2a), obtained from 2,6-dimethylpyridine, and iodide (3), obtained from a mixture of AgI/saturated KI in MeOH solutions, are isomorphous, monoclinic, P 21/c a≈ 24·2, b ≈ 13·9, c ≈ 20·2 Å, β ≈ 109·5°, Z = 4 dimers; R was 0·046 and 0·044 for No 5670 and 6039 respectively. The thiocyanate (4) has a similar cell, a 24·12(1), b 12·558(8), c 23·244(4) Å, β 110·11(3)°, Z = 4 dimers, R being 0·044 for No 7956; one of the thiocyanate ligands (which bridge in Ag-SCN-Ag mode) is disordered. A second polymorph of the bromide (2b) (from a mixture of AgBr/saturated KBr in H2O) is also monoclinic, P 21/c, a 14·121(8), b 25·577(3), c 21·968(2) Å, β 125·54(3)°, Z = 4 dimers (R was 0·047 for No 5715). Ag–As range between 2·568(1) and 2·633(1) Å throughout the series; in the isomorphous bromide and iodide, values increase slightly: 2·578(1)–2·611(1), cf. 2·601(2)-2·633(1) Å respectively. Ag–X are 2·568(2)-2·670(2) (Cl); 2·688(2)–2·715(2) (Br); 2·828(2)–2·856(1) Å (I); Ag-S, N for the ordered SCN group are 2·646(3), 2·255(6) Å. A redetermination of improved precision (R 0·035, No 6030) is reported for the triphenylphosphine/thiocyanate analogue. The far-infrared spectra of [(Ph3As)2Ag(µ-X)2Ag (AsPh3)2] show v(AgX) bands at 185, 145 (X = Cl), 145, 130, 106 (X = Br) and 121 cm-1 (X = I). The splittings and band widths reflect a decrease in the degree of distortion of the Ag(µ-X)2Ag units from a symmetrically bridged structure from X = Cl to I.

Photolysis of the N-chloroacetyl imide derivative 3-chloroacetyl-7,8-dimethoxy-4,5-dihydro-1H-3-benzazepin-2(3H)-one (2) in aqueous acetonitrile gave 11a-hydroxy-8,9-dimethoxy-5,6,11,11a-tetrahydro-oxazolo[2,3-b][3]benzazepin-3(2H)-one (4) in 81% yield. This representative of a new heterocyclic system could be converted into the 11a-ethoxy derivative (5) by recrystallization from ethanol. The structure of (5) was confirmed by a room-temperature single-crystal X-ray study. Crystals are monoclinic, Cc, a 17·623(9), b 7·550(4), c 12·872(3) Å, β 112·55(4)°, Z = 4; conventional R on F was 0·034 for 1072 ‘observed’ (I > 3σ(I)) reflections.

Abstract:

An LC-DAD-ESI-MSn technique was developed to investigate crude extracts of the stem bark of S. guineense for phenolic compounds. Ellagic acids were detected on the basis of their UV absorption spectra and ESI-MS fragmentation patterns. Six ellagic acid conjugates in the complex extract of S. guineense were identified. With the aim to confirm the structures, further purification of the extract afforded the known 3-O-ellagic acid-4′-O-α-rhampyranonosides (3), two new ellagic acid rhamnopyranosides, 3-O-methylellagic acid-4′-O-α-2″-O-acetylrhamnopyranoside (4) and 3-O-methylellagic acid-4′-O-α-3″-O-acetylrhamnopyranoside (6). The structures were identified on the basis of 1D- and 2D- NMR, IR, and UV spectroscopic and mass spectrometric data. The HMBC and NOESY data of these compounds were most useful for determination of their structures. Radical scavenging activities were evaluated in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Compounds 3, 4 and 6, with IC50 values of 6.1 ± 0.5, 5.8 ± 0.4, and 6.2 ± 0.4 μM, respectively were as active as ellagic acid (1) (IC50 = 4.9 ± 0.3 μM) and more active than catechin (IC50=10.8 ± 0.3 μM).

Abstract:

Shrimps are good source of protein and also good exportable item for its quality all over the world. Inappropriate use of antibiotics in the production of local and export quality shrimps can facilitate infiltration of antibiotic residues into human bodies which are toxic enough to cause serious health hazard. The present study was investigated to determine the antibiotic residues in some selected local market and export quality shrimps and also commercial shrimp feeds in chattogram. Local grade and export grade samples of Black tiger shrimps (Penaeus monodon) and feeds of four different companies were taken for the completion of this study. The permissible residual limit for the shrimp samples were- 0.3(μg/kg) for Chloramphenicol and 1.0 (μg/kg) for AHD, AMOZ, SEM, AOZ. Black tiger shrimps from local market 1 had high concentration of Chloramphenicol (0.35 μg/kg) and SEM (1.2 μg/kg) residues whereas Market 2 showed maximum residues in AMOZ (1.2 μg/kg) and SEM (1.1 μg/kg). Whereas, Market 3 showed negative result. Local market 4 had high concentration of Chloramphenicol (0.4 μg/kg) and AOZ (1.2 μg/kg) residues. However, in export grade shrimps antibiotic residues was found under the maximum residual limit in all samples. Also, among four feed tested, two of them were found positive for chlorampenicol and nitrofurans. The result was considered significant where P>0.05. So, legal actions should be taken against the farmers and the farms that overuse antibiotics in shrimps. Abstract:

Background: The treatment strategy for pediatric anterior cruciate ligament (ACL) tears, especially in patients with open physes, remains controversial. Purpose: To assess clinical outcomes and postoperative complications after all-epiphyseal double-bundle ACL (DB-ACL) reconstruction for patients with open physes. Study Design: Cohort study; Level of evidence, 3. Methods: Included were 102 patients aged ≤15 years who underwent ACL reconstruction at a single institution and had a minimum of 2 years of follow-up. Of these patients, 18 had undergone all-epiphyseal DB-ACL reconstruction (mean age, 12.4 ± 1.2 year) and 84 had undergone conventional DB-ACL reconstruction (mean age, 14.1 ± 0.9 year). The outcomes of the all-epiphyseal group were compared with those of the conventional group. Objective clinical outcomes included KT-1000 arthrometer measurements of side-to-side difference in anterior tibial translation, Lachman test grade, and pivot-shift test grade. Radiographic angular deformity (defined as >3° of the side-to-side difference in femorotibial angle) and incidence of second ACL injury were also compared. Results: The postoperative side-to-side difference in laxity significantly improved from 6.1 ± 2.4 to 0.6 ± 0.9 mm in the all-epiphyseal group ( P = .001), and postoperative laxity was similar to that of the conventional group (0.4 ± 0.8 mm; P = .518). A Lachman grade 1 positive result was observed in 20% of the all-epiphyseal group and 3% of the conventional group ( P = .042), and a pivot-shift grade 1 positive result was observed in 22% of the all-epiphyseal group and 4% of the conventional group ( P = .074). A total of 4 patients (26.7%) in the all-epiphyseal group and 4 (6.1%) in the conventional group demonstrated angular deformity ( P = .035). The incidence of postoperative ipsilateral ACL tear was 16.7% in the all-epiphyseal group and 23.8% in the conventional group ( P = .757). The incidence of contralateral ACL tear was 11.1% in the all-epiphyseal group and 14.3% in the conventional group ( P ≥ .999). Conclusion: All-epiphyseal DB-ACL reconstruction for skeletally immature patients achieved satisfactory clinical outcomes compared with conventional DB-ACL reconstruction. The incidence of ipsilateral graft rupture was relatively high in both groups. The all-epiphyseal group had a significantly higher incidence of angular deformity.

Abstract:

The aim of this study was to assess the epidemiological characteristics of maxillofacial trauma in the Piracicaba and Santa Barbara region - Sao Paulo, in a 4-year period. A descriptive and retrospective study was developed to analyze the epidemiological profile of facial trauma cases treated by the Residency Program of the Hospital of Sugarcane Suppliers of Piracicaba e Santa Barbara Hospital, through the evaluation of 625 medical records between March 2017 and December 2020, over a period of 04 years. Variables such as age, gender, etiology, type of trauma, type of treatment and presence or absence of postoperative complications were analyzed. The predominant age group was that of adults (20-59 years), the male gender was more affected with 76.74% of the sample. The most frequent etiology was a fall from a height (22.58%), followed by physical aggression (19.84%). Jaw fractures (17.07%) were more prevalent, followed by fractures of the nasal bone (16.26%). Conservative treatment of fractures was predominant (54.09%). The sample had a low complication rate (2.56%) and infections were the most observed complications (48.39%). The epidemiological characteristics of the population studied through data collection allowed the presentation of a profile of individuals affected by oral and maxillofacial trauma. The evaluation of the types of treatments performed and the complications allows a better perception of the particularity of the management of patients with facial trauma.

ImportanceDespite federal initiatives encouraging the enrollment of individuals from racial and ethnic minority groups in US clinical trials, no studies to date have specifically examined demographic disparities among participants in phase 1 drug development trials for patients with metastatic cancer.ObjectiveTo assess trends in the enrollment of patients from racial and ethnic minority groups in US phase 1 therapeutic drug trials for metastatic cancer from 2000 to 2018.Design, Setting, and ParticipantsIn this cross-sectional study, ClinicalTrials.gov was queried in July 2021 to identify completed phase 1 drug trials for metastatic cancer in the US from January 1, 2000, to December 31, 2018, with published results, yielding 221 phase 1 trials with 8309 participants aged 18 years or older with metastatic solid tumors. Proportions of each racial and ethnic group of trial participants were compared with that from the North American Association of Central Cancer Registries’ Cancer in North America (CiNA) database. Statistical analysis was performed from July 12, 2021, to March 15, 2022.Main Outcomes and MeasuresFor each racial and ethnic group, the difference between trial and CiNA proportions was examined using a 2-sample test for equality of proportions with continuity correction.ResultsThe 8309 phase 1 trial participants (4198 men [50.5%]; median age, 59 years) included 23 American Indian or Alaska Native participants (0.3%), 371 Asian or Pacific Islander participants (4.5%), 514 Black participants (6.2%), 401 of 5076 Hispanic or Latinx participants (7.9%), and 7154 White participants (86.1%). Industry funded 165 of the 221 trials (74.7%). White patients were overrepresented overall compared with the corresponding CiNA cohort (7154 of 8309 [86.1%] vs 4 113 096 of 4 891 486 [84.1%]; difference, 2.0 percentage points; P &amp;lt; .001). These findings may represent widening inequalities in access to trial sites and worsening systemic biases. More efforts are needed to diversify phase 1 cancer drug trials to improve equity in access to new treatments and to ensure that safety and efficacy findings from early drug trials are generalizable across populations.

Abstract:

We investigated whether two different bouts of high-intensity interval exercise (HIIE) could attenuate postprandial endothelial dysfunction. Thirteen young (27 ± 1 yr), nonexercise-trained men underwent three randomized conditions: 1) four 4-min intervals at 85–95% of maximum heart rate separated by 3 min of active recovery (HIIE 4 × 4), 2) 16 1-min intervals at 85–95% of maximum heart rate separated by 1 min of active recovery (HIIE 16 × 1), and 3) sedentary control. HIIE was performed in the afternoon, ~18 h before the morning fast food meal (1,250 kcal, 63g of fat). Brachial artery flow-mediated dilation (FMD) was performed before HIIE ( baseline 1), during fasting before meal ingestion ( baseline 2), and 30 min, 2 h, and 4 h postprandial. Capillary glucose and triglycerides were assessed at fasting, 30 min, 1 h, 2 h, and 4 h (triglycerides only). Both HIIE protocols increased fasting FMD compared with control (HIIE 4 × 4: 6.1 ± 0.4%, HIIE 16 × 1: 6.3 ± 0.5%, and control: 5.1 ± 0.4%, P < 0.001). For both HIIE protocols, FMD was reduced only at 30 min postprandial but never fell below baseline 1 or FMD during control at any time point. In contrast, control FMD decreased at 2 h (3.8 ± 0.4%, P < 0.001) and remained significantly lower than HIIE 4 × 4 and 16 × 1 at 2 and 4 h. Postprandial glucose and triglycerides were unaffected by HIIE. In conclusion, HIIE performed ~18 h before a high-energy fast food meal can attenuate but not entirely eliminate postprandial decreases in FMD. This effect is not dependent on reductions in postprandial lipemia or glycemia.NEW & NOTEWORTHY Two similar high-intensity interval exercise (HIIE) protocols performed ∼18 h before ingestion of a high-energy fast food meal attenuated but did not entirely eliminate postprandial endothelial dysfunction in young men largely by improving fasting endothelial function. Both HIIE protocols produced essentially identical results, suggesting high reproducibility of HIIE effects.

Abstract:

Background—The study aims to assess the efficacy of a permethrin 54.5%-fipronil 6.1%-based spot-on solution in dogs naturally infested with Neotrombicula in an open-label controlled study. Methods—Ten naturally infested dogs received one drop per affected site on day (D) 0, and on D14, the rest of the pipette was applied on the skin between the shoulders. Five dogs served as sentinels. Parasite score (0–3), skin lesions (0–4), and investigator pruritus scale (0–4) were assessed on D0, D1, D14, and D28. Results—No treated dogs developed adverse reactions. Parasite score of sentinel dogs was maintained between 1.8 (D0, D1, and D28) and 2.2 (D14). In treated dogs, D0 parasite score was 2.4. It was significantly reduced from D1 (0.5; p < 0.002) to D28 (0.1; p < 0.002). The lesion score was 2.9 on D0 and D1; it was significantly reduced on D14 (0.6; p < 0.002) and D28 (0.1; p < 0.002). Similarly, investigator pruritus scale (D0, 2.2) scores significantly decreased on D14 (0.4; p < 0.004) and D28 (0.2; p < 0.002). Conclusions—The combination permethrin-fipronil appears to be well-tolerated, rapidly and durably effective in the control of localized canine harvest mite infestation.

Abstract:

We demonstrate the concept of a spark sensor operating in UV range of the electromagnetic spectrum. The design of the device and the results of measurements of its most important characteristics are presented. Response time of the sensor is 3.8 ms and response threshold is 0.4 mW/m2. For the sensing element of the device glass material with the composition 10Li2O-15K2O-15Al2O3-35B2O3-25P2O5 doped with 5 at.% Eu is used. The peak of luminescence of the material corresponds to 611 nm when excited by a wide band (with wavelengths from 300 to 400 nm) of UV radiation that guarantees the high operation ability of the designed sensor.

Abstract:

The present research objective was to analyze the offensive phase from Complex I in high-level male volleyball teams in a macro- and micro-level view, through the inter e intra-team variability analysis of eight best teams of the 2018 Men’s Volleyball World Championship over the social network analysis and eigenvector centrality. The sample consisted of 22 matches and 2,743 offensive actions, resulting in 8 sub-networks with 368 nodes and 6221 edges. The results showed from macro view the variables that presented highest centrality values were Attack Zone 4 (range 0.56–0.90), Attack Tempo 2 (0.65–0.87), Power Attack (0.62–0.94), No Touch Block (0.61–1), Attack Effect Continuity (0.59–0.94), and Middle Blocker Centralized (0.60–0.95). In a micro view, Reception Effect, Play Position, Reception Zone, and Block Composition showed high variability in each sub-network. The intra- and inter-team variability presented the importance of to respect each team idiosyncrasies and to consider the different approaches to the game and success.

Abstract:

The present study examined the effects of the в-adrenergic receptor agonist, isoproterenol, on contractile activity of right ventricle papillary muscles in the rat heart at near-physiological temperature (30°C) and under conditions of deep hypothermia (10°C). Isoproterenol exerts a pronounced positive inotropic effect at 30°C. After agonist addition, the force of contraction increases from 1.2 ± 0.1 mN in control to 2.4 ± 0.4 mN that leads to a reliable acceleration of time parameters of contraction. So, time-to-maximum contraction decreased from 101 ± 6 ms to 85 ± 4 ms; time-to-50% relaxation declined from 55 ± 3 ms to 36 ± 1 ms. Under hypothermic conditions, isoproterenol produced a powerful negative inotropic effect, reducing the force of contraction from 2.2 ± 0.4 mN to 1.2 ± 0.4 mN. Similarly as at 30°C, there was a tendency for increase in contraction speed, so time-to-maximum contraction decreased from 717 ± 52 ms to 624 ± 50 ms, and time-to-50% relaxation was shortened from 667 ± 86 ms to 450 ± 40 ms. Thus, under conditions of deep hypothermia at 10°C, the isoproterenol-induced inotropy changes from positive to negative, while negative lusitropic effect remains clear.

Abstract:

Aim. Analysis of hospital and long-term results of carotid endarterectomy (CEA) in different periods of acute cerebrovascular event (ACVE). Materials and Methods. The given study was retrospective and was conducted using the method of patients sampling. In the period from 2010 to 2019, 1113 patients with ACVE in history who were later conducted CEA, were selected. Depending on the time interval between the last ACVE and CAE, all the patients were divided into 4 groups: the 1st group in the acutest period of ACVE (1-3 days) (n=24; 2.2%); the 2nd group in the acute period of ACVE (up to 28 days) (n=493; 44.3%); the 3rd group in the early rehabilitation period of ACVE (up to 6 months) (n=481; 43.2%); the 4th group in the late rehabilitation period of ACVE (up to 2 years) (n=115; 10.3%). The long-term period was 34.812.5 months. Results. In the hospitalization period of observation the following complications were found: lethal outcome ((group 1 0%; group 2 0.4% (n=2); group 3 0.2% (n=1); group 4 0%; р=0.16)); myocardial infarction ((group 1 0%; group 2 0.4% (n=2); group 3 0%; group 4 0.9% (n=1); р=0.35)); ACVE/transient ischemic attack (TIA), ((group 1 4.2% (n=1); group 2 0.4% (n=2); group 3 0.2% (n=1); group 4 0%; р1-2=0.01; р1-3=0.009; р1-4=0.01)). By the end of hospitalization period the composite endpoint consisting of lethal outcome + myocardial infarction + ACVE/TIA made in group 1 4.2% (n=1), in group 2 1.2% (n=6), in group 3 0.4% (n=2), in group 4 2.6% (n=3), р=0.08. Complications of the long-term follow-up period were: lethal outcome from all causes ((group 1 25% (n=6); group 2 5.5% (n=27); group 3 7.3% (n=35); group 4 14% (n=16); р1-2=0.002; р1-3=0.008; р2-4=0.012)); lethal outcome from cardiovascular causes ((group 1 4.2% (n=1); group 2 3.6% (n=18); group 3 4.8% (n=23); group 4 5.2% (n=6); р=0.79)), myocardial infarction ((group 1 12.5% (n=3); group 2 3.6% (n=18); group 3 5.4% (n=26); group 4 6.1% (n=7); р=0.15)), ACVE/TIA ((group 1 16.6% (n=4); group 2 6.3% (n=31); group 3 6% (n=29); group 4 11.3% (n=13); р=0.05)); composite endpoint including lethal outcome + myocardial infarction + ACVE/TIA ((group 1 54.2% (n=13); group 2 15.4% (n=76); group 3 18.7% (n=90); group 4 31.3% (n=36); р1-2=0.0001; р1-3=0.0001; р1-4=0.005; р2-4=0.0006; р3-4=0.012)). Conclusion. Application of CEA demonstrated effectiveness and safety in the acute and early rehabilitation period of ACVE.

Abstract:

Abstract Background In myelodysplastic syndromes (MDS), chronic myelomonocytic leukemia (CMML) and acute myeloid leukemia (AML) achievement of complete remission (CR) is a prerequisite for potential cure. In AML, CR/CR with incomplete recovery (CRi) is deemed the major outcome associated with improved overall survival (OS); patients (pts) without CR/CRi are considered non-responders, and hematologic improvement (HI) without (assessment of) bone marrow (BM) blast clearance is considered treatment (trt) failure. Achievement of CR may not be necessary for prolonged OS in pts treated with azacitidine (AZA) (Pleyer L, Annals Hematol 2014, 1825; Schuh AC, ASH 2015, P575). Outside of clinical trials, BM evaluations (BME) are only performed in ~50% of pts (Dinmohamed, Leuk Res 2015, 177) when either response or progression are obvious from peripheral blood (PB) values, or when pts are unable or unwilling to have BME. Aims To assess of the impact of response type on AZA trt outcomes in multivariate adjusted analyses (MVA). Methods 1441 pts included in the Austrian Azacitidine Registry were analyzed (NCT01595295). Data cut-off was 1 July 21. Marrow response was assessed for MDS/CMML and AML at each BME; HI was assessed on day 1 of each AZA cycle (Döhner H, Blood 2017, 424; Cheson BD, Blood 2006, 419; Pleyer L, ASH 2019, P3821); peripheral blood complete remission (PB-CR) was defined as hemoglobin ³11 g/dL, platelet count ≥100 G/L, neutrophil count ³1.0 G/L, white blood cell count &lt;15 G/L, PB blasts =0%, and no transfusions. Response types were calculated from electronic case report form data. To identify which response type achieved by which AZA cycle had the highest impact on time-to-event endpoints, likelihood ratios (LR) of the Cox-regression model for OS or time to next treatment (TTNT) were calculated using the respective response types as covariates. Baseline characteristics with univariate p&lt;0·10 (n=23) for association with OS were included in the multivariate regression. After stepwise selection n=17 variables remained and were used for MVA. Assign Data Management and Biostatistics GmbH performed statistical analyses with SAS® 9.4. Results In total, 521, 135, and 785 pts had MDS, CMML and AML. Median year of initial diagnosis was 2012, median time to AZA start was 3·0 (IQR 1·0-13·2) months (mo), median follow-up time from AZA start was 10.6 (IQR 4·0-21·1) mo, 894 pts received AZA as first line trt, median age at AZA start was 73 (range 23-99) years. In total, 13956 AZA cycles were applied, median duration of AZA trt was 5·0 (IQR 1·9-12·1) mo, median AZA dose was 875 (IQR 700-1000) mg/cycle, AZA was applied for a median of 7 (IQR 5-7) days. Median time to best response was 3·7 (IQR 2·0-5·9) months. Early mortality was 5.5% within 30 days. During AZA trt 1225 BM evaluations (BME) were performed in 697 (48·4%) of pts. Of these, 204 achieved CR/CRi. Irrespective of BME, 622 (43%) of 1441 pts achieved an HI and 264 (18·3%) of 1441 pts achieved a PB-CR. Pts achieving CR had longer adjusted OS (23·7 vs 19·7 mo, p=0·0227; HR=0·621 [0·413-0·936]) and TTNT (19·4 vs 15·7 mo, p=0·0262; HR=0·644 [0·436-0·949]) than pts achieving CRi. Among pts achieving CR, those additionally achieving PB-CR had longer adjusted OS (24·8 vs 16·3; p=0·0040; HR=0·256 [0·101-0·647]; Fig 1A) and TTNT (21·2 vs 11·0; p=0·0005; HR=0·219 [0·094-0·513]; Fig 1B) than those who did not. Among pts not achieving CR, those additionally achieving PB-CR had longer adjusted OS (20·8 vs 14·1; p&lt;0·0001; HR=0·510 [0·397-0·657]; Fig 1A) and TTNT (17·7 vs 10·9; p&lt;0·0001; HR=0·485 [0·357-0·589]; Fig 1B) than those who did not. Among all pts, irrespective of BM blast count, achievement of PB-CR resulted in longer adjusted OS (21·7 vs 10·0 mo; p&lt;·0001; HR 0·363; Fig 1C) and TTNT (18.5 vs 7.8 mo; p&lt;0.0001; HR=0.346; Fig 1D) and provided added value to CR and CR/CRi. Among all response types and after MVA, the highest prognostic impact on both OS and TTNT was observed when achieving PB-CR or CR/CRi by cycle 9 or 10 (Fig 2A-B). Conclusions Above data indicate that achievement of PB-CR is a strong predictor of OS and TTNT that provides additional information to current response criteria. Inclusion of PB-CR in updated response criteria of pts with MDS, CMML or AML receiving non-intensive trt should be considered. The greatest advantage of PB-CR is that it can be easily, nearly painlessly and quickly assessed. Abstract:

The 650–621 Ma plume which impinged beneath the Siberian craton during the breakup of Rodinia caused the formation of several alkaline carbonatite massifs in craton margins of the Angara rift system. The Beloziminsky alkaline ultramafic carbonatite massif (BZM) in the Urik-Iya graben includes alnöites, phlogopite carbonatites and aillikites. The Yuzhnaya pipe (YuP) ~ 645 Ma and the 640–621 Ma aillikites in BZM, dated by 40Ar/39Ar, contain xenoliths of carbonated sulfide-bearing dunites, xenocrysts of olivines, Cr-diopsides, Cr-phlogopites, Cr-spinels (P ~ 4–2 GPa and T ~ 800–1250 °C) and xenocrysts of augites with elevated HFSE, U, Th. Al-augites and kaersutites fractionated from T ~ 1100–700 °C along the 90 mW/m2 geotherm. Higher T trend for Al-Ti augite, pargasites, Ti-biotites series (0.4–1.5 GPa) relate to intermediate magma chambers near the Moho and in the crust. Silicate xenocrysts show Zr-Hf, Ta-Nb peaks and correspond to carbonate-rich magma fractionation that possibly supplied the massif. Aillikites contain olivines, rare Cr-diopsides and oxides. The serpentinites are barren, fragments of ore-bearing Phl carbonatites contain perovskites, Ta-niobates, zircons, thorites, polymetallic sulphides and Ta-Mn-Nb-rich magnetites, ilmenites and Ta-Nb oxides. The aillikites are divided by bulk rock and trace elements into seven groups with varying HFSE and LILE due to different incorporation of carbonatites and related rocks. Apatites and perovskites reveal remarkably high LREE levels. Aillikites were generated by 1%–0.5% melting of the highly metasomatized mantle with ilmenite, perovskite apatite, sulfides and mica, enriched by subduction-related melts and fluids rich in LILE and HFSE. Additional silicate crystal fractionation increased the trace element concentrations. The carbonate-silicate P-bearing magmas may have produced the concentration of the ore components and HFSE in the essentially carbonatitic melts after liquid immiscibility in the final stage. The mechanical enrichment of aillikites in ore and trace element-bearing minerals was due to mixture with captured solid carbonatites after intrusion in the massif.

Abstract:

Egy műtrágyázási tartamkísérlet 28. évében, 2001-ben vizsgáltuk az eltérő N, P, K ellátottsági szintek és kombinációik hatását a réti csenkesz (Festuca pratensis) vezérnövényű, nyolckomponensű pillangós nélküli gyepkeverék ásványi elemfelvételére. A termőhely talaja a szántott rétegben mintegy 3% humuszt, 3-5% CaCO3-ot és 20-22% agyagot tartalmazott, N és K elemekben közepesen, P és Zn elemekben gyengén ellátottnak minősült. A kísérlet 4N x 4P x 4K = 64 kezelést x 2 ismétlést = 128 parcellát foglalt magában. A talajvíz 13-15 m mélyen helyezkedik el, a terület aszályérzékeny. A vizsgált 2001. évben kielégítő, 621 mm csapadék hullott és annak eloszlása is kedvező volt. A kísérlet módszerét, körülményeit, valamint a műtrágyázás termésre és a széna összetételére gyakorolt hatását előző közleményeink taglalták (Kádár, 2005, 2005a; Kádár és Győri, 2005). Főbb következtetések: 1. Az NxP pozitív kölcsönhatások eredményeképpen és a K-kezelések átlagában az egyes elemek felvétele az 1. kaszáláskor alábbi intervallumban ingadozott az N0P0 kontroll és a maximális N3P3 kínálat között: K 62-190, N 45-218, Ca 16-51, S 5-24, P 4-24, Mg 4-16, Na 0,5-5,0 kg/ha; Mn 282-968, Sr 35-170, Zn 32-73, Ba 29-55, B 18-44, Cu 8-40 g/ha. Hasonlóképpen a KxP pozitív kölcsönhatások nyomán a Ba 23-62, Ni 1,5-8,9 g/ha tartományban változott. A KxP negatív kölcsönhatások viszont K0P0 kontrollon mért 1,6 g/ha Mo felvételt 0,4 g/ha-ra mérsékelték. 2. A 2001. október 9-én történt 2. kaszálás idején a N-hatások érvényesültek: szénahozam a kontrollon mért 1,0 t/ha-ról 3,9 t/ha-ra nőtt. Ugyanitt a Fe, Ba és Mo felvétele kétszeresére; a Ca, S, P, Sr, Zn és Co felvétele 3-4-szeresére; a K, N, Mg, Mn, Ba, Cu és Ni felvétele 5-6-szorosára, míg a Na felvétele 33-szorosára nőtt a maximális N-kínálattal, a kontrollhoz viszonyítva a PK kezelések átlagában. 3. A két kaszálás összegeit tekintve a szénahozam a 28 éve trágyázatlan talajon 3 t/ha, míg a maximális N3P3K3 kezelésben 13 t/ha volt. Ugyanitt a felvett Fe, Cr, B, Ni, Mo és Co 2-3-szorosára; a Ca, Mg, Mn, K, Zn, Ba és Cu 5-6-szorosára; a S, Sr és P 7-8-szorosára; a N 10-szeresére, míg a Na 16-szorosára emelkedett. A kivont K és N maximális tömege elérte a 388 kg/ha, Ca 80 kg/ha, S 49 kg/ha, P 42 kg/ha (96 kg/ha P2O5), Mg 24 kg/ha mennyiséget. 4. Az 1 t széna képzéséhez szükséges átlagos fajlagos elemigény kísérleti viszonyaink között az alábbinak adódott: 25 kg K (30 kg K2O), 20 kg N, 6 kg Ca (8-9 kg CaO), 2-3 kg S, 2 kg P (5 kg P2O5), 2 kg Mg (3-4 kg MgO). Abstract:

The ( Li 0.5 Bi 0.5)( W 1-x Mo x) O 4(0.0 ≤ x ≤ 1.0) ceramics were prepared via the solid state reaction method. The sintering temperature decreased almost linearly from 755°C for ( Li 0.5 Bi 0.5) WO 4 to 560°C for ( Li 0.5 Bi 0.5) MoO 4. When the x≤0.3, a wolframite solid solution can be formed. For x = 0.4 and x = 0.6 compositions, both the wolframite and scheelite phases can be formed from the X-ray diffraction analysis, while two different kinds of grains can be revealed from the scanning electron microscopy and energy-dispersive X-ray spectrometer results. High performance of microwave dielectric properties were obtained in the (Li0.5Bi0.5)(W0.6Mo0.4)O4 ceramic sintered at 620°C with a relative permittivity of 31.5, a Qf value of 8500 GHz (at 8.2 GHz), and a temperature coefficient value of +20 ppm/°C. Complex dielectric spectra of pure (Li0.5Bi0.5)WO4 ceramic gained from the infrared spectra were extrapolated down to microwave range, and they were in good agreement with the measured values. The ( Li0.5Bi0.5 )( W 1-x Mo x) O 4(0.0 ≤ x ≤ 1.0) ceramics might be promising for low temperature co-fired ceramic technology.

Abstract:

Hydrolysis/[3 + 2] cycloaddition/elimination cascades employed for the synthesis of unexpected tricyclic compound derived from isoquinoline. Reaction of ethylene derivative 1 with the isoquinoline ester iminium ion 2 in alkaline medium (MeOH/NEt3) under reflux for 1 h resulted in the formation of the fused pyrrolo[2,1-a]isoquinoline derivative 3. Its structure was elucidated by X-ray single crystal and other spectrophotometric tools. Hirshfeld calculations for 3 and its crystal structure analysis revealed the importance of the short O…H (19.1%) contacts and the relatively long H…C (17.1%), Cl…H (10.6%) and C…C (6.1%) interactions in the molecular packing. DFT calculations were used to compute the electronic and spectroscopic properties of the studied system. The studied compound has polar nature (3.5953 Debye). TD-DFT calculations assigned the shortest wavelength band (220 nm) to the hom*o−1→LUMO+2 (57%), hom*o−1→LUMO+4 (14%) mixed excitations. The calculated NMR chemical shifts correlated very well with the experimental data (R2 = 0.93–0.94).

Abstract:

Abstract Background: Alpelisib and fulvestrant are used as a combination treatment option for postmenopausal PIK3CA-mutated, hormone receptor positive (HR+), human epidermal growth factor receptor 2-negative (HER2-), advanced or metastatic breast cancer (a/mBC) patients. However, despite the presence of activating mutations in PIK3CA, the majority of patients do not derive benefit, or ultimately progress while on alpelisib therapy. Here, we investigate the genomic landscape of PIK3CA-mutated, HER2- a/mBC using next-generation sequencing (NGS) to provide insight into possible mechanisms of therapeutic resistance to alpelisib/fulvestrant and to identify potential targetable pathways. Methods: We utilized the Tempus LENS platform to retrospectively analyze de-identified NGS data from 2,918 a/mBC patients with formalin-fixed, paraffin-embedded tumor biopsies sequenced using the Tempus|xT solid tumor assay (DNA-seq of 595-648 genes at 500x coverage; full transcriptome RNA-seq). Mutations identified included germline and/or somatic single nucleotide variants, insertions/deletions and copy number variations (gains defined as ≥8 copies). We used curated clinical data to determine HER2 and hormone receptor (ER/PR) status. Results: Among 2,918 a/mBC patients, we identified somatic mutations in PIK3CA in 782 (26.8%). Within these tumors, 629 (80.4%) had one of the 11 mutations currently included in the alpelisib companion diagnostic, and we focused on this population (here defined as mut-PIK3CA). Of these 629, 546 (86.8%) were HER2-, with 176 (32.3%) and 370 (67.7%) derived from primary and metastatic tumors, respectively. Cases were further classified as HR+ (defined as ER+ or PR+) or triple negative (TNBC). While the majority of mutPIK3CA samples were identified in HR+ disease, 10% of the cases occurred in TNBC. Within the mutPIK3CA cohort, tumor mutational burden high (TMB-H; defined as ≥10 mutations/MB) was detected in 11.5% of samples, while microsatellite instability high (MSI-H) was detected in 0.5%. MSI-H was detected at a higher frequency in TNBC compared to HR+. Overall, the most commonly co-mutated genes among mutPIK3CA, HER2- samples were TP53 (34.6%), CDH1 (21.6%), ESR1 (12.3%), KMT2C (11%), MAP3K1 (9.5%), ARID1A (8.1%), PTEN (6.8%), GATA3 (6.6%), NF1 (5.9%), and TBX3 (5.9%) among others (Table 1); some of these genes have previously been implicated in resistance to endocrine therapy or PI3K inhibitor. In addition, in HR+ disease, metastatic samples had a higher frequency of mutations in genes implicated in endocrine resistance, such as ESR1 (18.7% vs 1.9%), ERBB2 (3.3% vs 2.6%), NF1 (6.8% vs 2.6%), compared to primary tumors. We also identified copy number gains (CNG) in several cell cycle genes, including: CCND1 (15.2%), CDK4 (2.7%), and AURKA (2.6%) (Table 1). Finally, further analyses at the transcript-level are the subject of on-going research. Conclusions: Our study highlights that there is substantial genomic heterogeneity among mutPIK3CA, HER2- a/mBCs. Across a series of comparisons between primary and metastatic samples, as well as HR+ and TNBC subtypes, we identified a number of co-mutations that occur alongside mutPIK3CA and which could be potentially exploited by targeted therapies. Future studies are needed to assess the prognostic/predictive role of these and other candidate gene alterations. Table 1. Abstract:

Abstract Background Fibrosis constitute the main complications associated to Crohn’s disease (CD). Notch signalling has been implicated in lung, kidney, liver and cardiac fibrosis. Macrophages contribute to fibrosis through the release of different mediators and the pattern of secretion may vary according to their microenvironment. The aim of the present study is to analyze the role of Notch ligands derived from macrophages in the complications of CD. Methods We have analyzed: the mRNA expression of cytokines and Notch ligands in CD patients with fistulizing and stenting pattern, the mRNA and protein expression of macrophage markers and Notch ligands in macrophages treated with the main cytokines present in CD patients (IFNγ-, IL10-, IL4, TNFα-U937 treated cells), the mRNA expression of fibrosis markers of DLL4-HT29 treated cells. Results are expressed as fold induction (mean±SEM, n≥4). Statistical analysis was performed with one-way ANOVA followed by Newman-Keuls test. Correlations were analyzed with the Spearman coefficient. Results The mRNA expression of IFNγ, TNFα, IL4, IL6 and IL10 was significantly higher in intestinal samples from B2- and B3-CD patients (11.4±1.6, 6.9±1.3, 16.4±3.9, 4.1±0.4, and 5.0±1.0 respectively for B2, and 14.2±1.8, 9.6±2.6, 21.5±4.1, 6.3±1.0 and 9.2±1.7 respectively for B3) than in controls (1.0±0.09, 1.1±0.2, 1.2±0.1, 1.1±0.1 and 1.0±0.1 respectively). The mRNA expression of DLL3 and DLL4 was significantly higher in intestinal samples from B2 (6.1±1.3 and 9.3±2.4, respectively) than in B3 CD patients (4.4±1.0 and 1.8±0.7, respectively) and in controls (0.9±0.2 and 0.7±1.2 respectively). IFNγ-U937 treated cells increased significantly the mRNA expression of DLL3 and DLL4 (2,9±0,6 N=5 N=4 and 3,7±1,1 N=4, respectively) respect vehicle; IL1β increased significantly the expression of DLL4 (1,8±0,01 N=4) and TNFα increased significantly the expression of DLL3 (7,1±2,2 N=4), respect vehicle. DLL4-HT29 treated cells increased significantly fibrosis markers (VIMENTIN (4,6±0,6 N=6), α-SMA (20,9±8,2 N=6), SNAIL1 (1,8±0,4 N=6), SNAIL2 (8,3±1,3 N=6), ZEB1 (8,2±4,3 N=6) and ZEB2 (4,3±0,2 N=6), respect vehicle. Conclusion Macrophages may act as a source of Notch ligands who could act as fibrosis mediators in CD patients with a stenting (B2) behavior. The microenvironment rich in IFNγ could activate the fibrosis process in epithelial cells by favoring the expression of DLL4 in macrophages.

Abstract:

In Beetal goats under subtropical conditions, ovulatory response of first-wave dominant follicle based on diameter and day of oestrous cycle is unknown to date. The objective of this study was to evaluate the ovulatory response to gonadotropin-releasing hormone (GnRH) relative to day and diameter of dominant follicle during the first follicular wave in Beetal goats. Fifty goats were synchronized using a single dose of prostaglandin F2α (75 µg; D+ cloprostenol, Fatro, Italy), and scanned every 6 hours for ovulation using a 7.5-MHz trans-rectal transducer (Honda HS: 1500, Japan). Following ovulation (Day 0), goats (n=40; 10/day) were given GnRH (25µg lecirelin, Fatro, Italy) on Days 2, 4, 6, and 8 of oestrous cycle. Follicular dynamics was monitored daily from D 0 until GnRH, and every 4 hours after GnRH until ovulation. Ovulation frequency relative to day and dominant follicular diameter was analysed by binary logistic regression, whereas diameter of dominant follicle at time of GnRH, ovulation time, time of wave emergence and growth rate of dominant follicle following GnRH were analysed by one-way ANOVA and independent sample t-test (SPSS 20, IBM Corp., Armonk, NY, USA). Eighty percent of the goats ovulated within 74.7±1.3h after prostaglandin F2α. Overall, 37% (14/38) of the goats ovulated following GnRH within 12.0±1.5h. Ovulations (%) following GnRH were higher (P=0.03) on Day 4 than 2 (60 v. 10%; i.e. 14 times higher than Day 2) and decreased gradually from Day 4 to 8 (P&gt;0.05). Similarly, ovulations following GnRH were higher (P=0.00) in first-wave dominant follicles having diameter 6.1-7.0mm than those having less than 6.1mm (88v. 23%; i.e. 24 times higher than &lt;6.1mm). Ovulations (%) decreased as the diameter of dominant follicle increased from 6.1-7.0mm (P&gt;0.05). Diameters of ovulatory follicles at time of GnRH between Day 2, 4, 6, and 8 were not different (P&gt;0.05). Regardless of day of GnRH treatment, diameter of ovulatory v. anovulatory follicles differed (6.44±0.03v. 5.1±0.4 mm; P=0.00). Overall, growth rate of dominant follicles following GnRH was 0.2±0.11mm and did not differ between ovulatory and anovulatory goats (0.29±0.13v. 0.13±0.15 mm; P&gt;0.05). Interval to the emergence of a new wave following GnRH was prolonged for Day 4 compared to Day 2, 6, and 8 (i.e. 22v. 8, 11, and 12 h; respectively, P&lt;0.05). In conclusion, GnRH treatment at Day 4 resulted in maximum ovulations as the first-wave dominant follicle reached a size of 6.1 to 7.0 mm.

Os tratamentos resultaram da combinação entre dois fatores: cinco níveis de condutividade elétrica da água de irrigação - CEa (0,4; 3,1; 4,1; 6,1 e 7,1 dS m-1) associados a inoculação ou não das sem*ntes de algodão com Azospirillum brasilense - AZ (Com inoculação e sem inoculação), distribuídos no delineamento inteiramente casualizado, em arranjo fatorial 5 × 2, com seis repetições e uma planta por parcela, perfazendo o total de sessenta unidades experimentais. A inoculação das sem*ntes com Azospirillum brasilense mitigou os efeitos deletérios do estresse salino sobre a área foliar e os componentes de produção quando submetido a CEa de até 3,1 dS m-1. The treatments resulted from the combination of two factors: five levels of electrical conductivity of the irrigation water - ECw (0.4, 3.1, 4.1, 6.1 and 7.1 dS m-1) associated with inoculation or not of cotton seeds with Azospirillum brasilense - AZ (With inoculation and without inoculation), distributed in a completely randomized design, in a 5 × 2 factorial arrangement, with six repetitions and one plant per installment, making a total of sixty experimental units. Seed inoculation with Azospirillum brasilense mitigated the deleterious effects of saline stress on leaf area and yield components when subjected to ECw of up to 3.1 dS m-1. Irrigation with ECw of 0.4 dS m-1 associated with seed inoculation with Azospirillum brasilense resulted in the highest values ​​of leaf area, shoot dry biomass, boll number, seed number, seed weight and cotton weight in feather. This ongoing first-in-human study assesses safety, biologic, and clinical activity of BPX-601+Rim in PSCA+ cancers. Updated results, including those from patients (pts) who underwent LD with Flu/Cy, are presented. Methods: BP-012 is a 2-part, open-label trial. Part 1 is a 3+3 dose escalation of BPX-601 (1.25–5.0x106 cells/kg; Day [D] 0) given prior to a single, fixed Rim dose (0.4 mg/kg; D7) in pts with previously treated PSCA+ metastatic pancreatic, gastric, or prostate cancers with measurable disease. Results: As of Jan-22-2019, 15 pts have received BPX-601±Rim. Two pts at the highest cell dose received Flu/Cy for LD on D−5 to D−3 before BPX-601; LD after Flu/Cy was 96.6% and 84.3%. Thirteen pts received Cy alone on D−3; in these pts, LD ranged from 0–68.6%. Rapid cell expansion by D4 was observed in all pts with peak vector copy number 8.3-fold higher with Flu/Cy (n = 2) vs Cy LD (n = 13). Serum IP-10, IL-6 and TNFα increased > 2-fold from baseline in ≥1 pt in all Rim cohorts, with 3- to 20-fold Rim-dependent cell expansion in 6 pts. No CRS or DLTs were reported. After Rim, one Flu/Cy pt experienced a serious Grade 2 AE (encephalopathy) related to BPX-601+Rim that resolved with IV steroids; despite time-matched nonserious Grade 1 pyrexia, the pt had no other CRS symptoms. After BPX-601+Rim and ≥1 scan, best responses were 8 SD and 3 PD (1 non-evaluable). With a median follow-up of 9.8 wks, time to next treatment (tx) after BPX-601 ranged from 2.7–22.1 wks (n = 8) and ongoing tx-free intervals range from 9.1–30.1 wks (n = 4). Conclusions: BPX-601+Rim was well-tolerated with manageable safety and early evidence of enhanced CAR T-cell expansion and prolonged persistence after Flu/Cy vs Cy. Additional pts will undergo Flu/Cy LD prior to BPX-601 with single- and repeat-dose Rim. Clinical trial information: NCT02744287.

Abstract:

Abstract. The impact of aerosol composition on cloud condensation nuclei (CCN) activity was analyzed in this study based on field experiments carried out at downtown Tianjin, China, in September 2010. In the experiments, the CCN measurements were performed at supersaturation (SS) of 0.1%, 0.2% and 0.4% using a thermal-gradient diffusion chamber (DMT CCNC), whereas the aerosol size distribution and composition were simultaneously measured with a TSI SMPS and an Aerodyne Aerosol Mass Spectrometer (AMS), respectively. The results show that the influence of aerosol composition on CCN activity is notable under low SS (0.1%), and their influence decreased with increasing SS. For example, under SS of 0.1%, the CCN activity increases from 4.5 ± 2.6% to 12.8 ± 6.1% when organics fraction decrease from 30–40% to 10–20%. The rate of increase reaches up to 184%. While under SS of 0.4%, the CCN activity increases only from 35.7 ± 19.0% to 46.5 ± 12.3%, correspondingly. The calculated NCCN based on the size-resolved activation ratio and aerosol number size distribution correlates well with observed NCCN at high SS (0.4%), but this correlation decreases with the falling of SS. The slopes of linear fitted lines between calculated and observed NCCN are 0.708, 0.947, and 0.995 at SS of 0.1%, 0.2% and 0.4%, respectively. Moreover, the standard deviation (SD) of calculated NCCN increases with the decreasing of SS. A case study of CCN closure analyses indicates that the calculated error of NCCN can reach up to 34% at SS of 0.1% if aerosol composition is not included, and the calculated error decreases with the raising of SS. It decreases to 9% at SS of 0.2%, and further decreases to 4% at SS of 0.4%.

Abstract:

BackgroundCarbon dioxide (CO2) is a pervasive chemical stimulus that plays a critical role in insect life, eliciting behavioral and physiological responses across different species. High CO2concentration is a major feature of termite nests, which may be used as a cue for locating their nests. Termites also survive under an elevated CO2concentration. However, the mechanism by which elevated CO2concentration influences gene expression in termites is poorly understood.MethodsTo gain a better understanding of the molecular basis involved in the adaptation to CO2concentration, a transcriptome ofCoptotermes formosanusShiraki was constructed to assemble the reference genes, followed by comparative transcriptomic analyses across different CO2concentration (0.04%, 0.4%, 4% and 40%) treatments.Results(1) Based on a high throughput sequencing platform, we obtained approximately 20 GB of clean data and revealed 189,421 unigenes, with a mean length and an N50 length of 629 bp and 974 bp, respectively. (2) The transcriptomic response ofC. formosanusto elevated CO2levels presented discontinuous changes. Comparative analysis of the transcriptomes revealed 2,936 genes regulated among 0.04%, 0.4%, 4% and 40% CO2concentration treatments, 909 genes derived from termites and 2,027 from gut symbionts. Genes derived from termites appears selectively activated under 4% CO2level. In 40% CO2level, most of the down-regulated genes were derived from symbionts. (3) Through similarity searches to data from other species, a number of protein sequences putatively involved in chemosensory reception were identified and characterized inC. formosanus, including odorant receptors, gustatory receptors, ionotropic receptors, odorant binding proteins, and chemosensory proteins.DiscussionWe found that most genes associated with carbohydrate metabolism, energy metabolism, and genetic information processing were regulated under different CO2concentrations. Results suggested that termites adapt to ∼4% CO2level and their gut symbionts may be killed under high CO2level. We anticipate that our findings provide insights into the transcriptome dynamics of CO2responses in termites and form the basis to gain a better understanding of regulatory networks.
